Serotonin (5-hydroxytryptamine, 5-HT), a monoamine neurotransmitter, is biochemically derived from tryptophan. 5-HT is primarily found in gastrointestinal (GI) tract, platelets, and in central nervous system (CNS). Approximately 90% of total serotonin in human body is located in enterochromaffin cells of the alimentary canal and 10% is synthesized in serotonergic neurons of CNS, a place where it has been implicated in various functions. Serotonin is mainly metabolized to 5-HIAA. The first line of action in its metabolism is oxidation by monoamine oxidase which converts it to the corresponding aldehyde. Disturbance of serotonin homeostasis in the body leads to diverse physiologic and pathologic changes, including the presence of clinical diseases. This chapter reviews the possible mechanisms of 5-HT and their metabolites that are involved in usual clinical disorders in CNS. In addition, the analytical techniques and biochemical markers used in monitoring 5-HT and their metabolites in the same disorders which have been used in several investigations reported in our lab are also described in this recompilation. © 2013 Nova Science Publishers, Inc.
