Resumen:

Metabolic control analysis (MCA) has shown that control of pathways and cellular processes is distributed among several participants and, hence, there are not single rate-limiting steps. In this article, four cases are reviewed in which the MCA framework provides tools for the identification of the best targets for therapeutic intervention in cancer and parasitic diseases, and rationalizes why some selected trial-and-error strategies to overproduce metabolites of biotechnological interest have been successful. © 2019 Elsevier B.V.
