Resumen:

Transforming growth factor (TGF)-ß is a key regulator of cell proliferation, survival, and differentiation, controlling bone development, and homeostasis. Perturbations of TGF-ß signaling result in the development of tumors that can affect the skeleton. Although TGF-ß is a tumor suppressor, cancer cells can subvert the TGF-ß signaling pathway into a tumor and metastasis promoter. In this chapter, we will review the function of the TGF-ß signaling pathways in multiple skeletal complications of malignancy, including multiple myeloma, different primary cancer of the bone, and bone metastases, and how TGF-ß affects cancer cells, the tumor microenvironment, and distant organs. As the molecular and cellular mechanisms are becoming increasingly clear, they emphasize the potential of TGF-ß as a therapeutic target. However, the promising results in preclinical models are not translated to the clinic so far. We will highlight how combination of anti-TGF-ß treatment with other therapies could improve the outcome of cancer patients. © 2022 Elsevier Inc. All rights reserved.
