The generalized understanding of the mechanisms of ion channels at the level of a single molecule was made possible by the development of a robust technique known as patch-clamp recording. This application is an important subset of the voltage clamp, and as such, permits exquisite control of the membrane potential difference and the added advantage of being able to control the composition of solutions bathing the cell membrane and the ion channels in it. While relatively straight forward in its application, patch-clamp recordings are so sensitive that they are subjected to potential artifacts and a thorough knowledge of the phenomena involved in its application is needed. The intensive use of patch clamp for over four decades now has made possible the detailed understanding of the biophysics of ion channels and continues to be developed. This chapter discusses the fundamentals of patch clamp and the analysis of resulting single-channel records necessary to derive insights on ion channel mechanisms. © 2023 selection and editorial matter, Jie Zheng and Matthew C.
