© 2025 Elsevier Inc. All rights reserved.The subject of this chapter is pyroglutamyl-peptidase II, a membrane-bound metallopeptidase that releases an N-terminal pyroglutamyl residue from a peptide. Because pyroglutamyl is an amino acid derivative, pyroglutamyl-peptidase II is more properly classified as an omega-peptidase rather than an aminopeptidase. Its physiological substrates include thyrotropin-releasing hormone, which is inactivated by the release of the N-terminal pyroglutamate. Pyroglutamyl-peptidase II is a Glu-zincin and a member of family M1. It is a type II integral membrane protein located on the surface of neuronal and pituitary cells and tanycytes.
