All rights reserved.Raman imaging combined with Principal Component Analysis (PCA) was employed to investigate the vibrational morphology of three cell lines: two breast cancer cell lines and one control. Raman imaging provides spatially resolved molecular information, allowing the identification of distinct cellular regions based on their biochemical composition. Live cells were analyzed using a confocal Raman microscope, acquiring Raman spectra from individual pixels to reconstruct high-resolution vibrational maps. PCA was applied to extract the most relevant spectral variations, enabling the differentiation of intracellular zones such as the nucleus, cytoskeleton, and extracellular region. The vibrational fingerprints associated with these regions revealed key molecular differences, including nucleic acid signatures in the nucleus and cytoskeletal protein features in the cytoplasm. The results demonstrate the potential of Raman imaging as a label-free, non-invasive method for cellular characterization, offering precise molecular insights under near-physiological conditions. These findings highlight the applicability of Raman imaging in biomedical research, diagnostics, and translational medicine.
